MACRO- AND MICRO ELEMENTS IN TISSUES OF SOME SPECIES OF ASCIDIANS OF PETER THE GREAT BAY (SEA OF JAPAN)

Abstract. Chemical composition of tunic and cloak of an ascidian Styela clava, Boltenia echinata, and Ciona intestinalis of Peter the Great Bay (Sea of Japan) is studied. Substantial specific differences are educed in an accumulation mаcro- and microelements by a tunic and cloak of the indicated spesies of ascidias. Level of maintenance of elements at Boltenia echinata on the whole in several times is higher, that at Styela clava, concentration of sulphur, calcium and iron arrives at the most high values. At Ciona intestinalis the concentration of microelements considerably below as compared to other types of ascidias, abromine and iron prevail in tissues.

Keywords: macroelements, microelements, tunic, clock, ascidian.

Abstract. The maintenance of some macro- and microelements in muscle tissue, skin, gills, gonads and liver of flounder H. dubius were studied. In liver the maximum concentrations of iron, copper, selenium, in gonads – zinc, in bone tissue – manganese, in the gills – bromine were detected. Chromium is found only in the skin and muscle tissue of this species. The concentration of As, Zn, Cu, Cd in the organs of H. dubius does not exceed the maximum possible concentration.

Keywords: microelements, flounder southern halibut H. dubius, liver, skin, gills, gonad, bone, muscles.

ONE-MONTH ALTERATION IN THE ANTIOXIDANT DEFENSE IN THE HEART OF RAINBOW TROUT (ONCORHYNCHUS MYKISS WALBAUM) VACCINATED AGAINST YERSINIA RUCKERI

Abstract. The present work aimed to assess the effects of oral vaccination against Yersinia ruckeri at first month after immunization in the cardiac tissue of rainbow trout (Oncorhynchus mykiss Walbaum). Concentrated vaccine with inactivated by formalin Y. ruckeri strains was enclosed by fish feed, and was administered three times every other day. Our results suggest that antioxidant reaction rates, characteristic of immunization, may be sufficient to protect against at least some of the physiological changes that might promote oxidative stress. In addition, our observation that products of lipid peroxidation are higher in the cardiac tissue from vaccinated animals (but not oxidatively modified proteins) indicates that antioxidant defenses is important in setting the balance in oxidative stress during immunization. Correlations between aldehydic derivatives of oxidatively modified proteins and antioxidant defenses confirmed that increased superoxide dismutase activity caused to decreased level of protein damage in the cardiac tissue of the trout vaccinated against Y. ruckeri at first month after immunization. Discrepancies observed for antioxidant enzymes among vaccinated fishes following oral vaccination against Y. ruckeri may represent tissue specificvariation required to support tissue-specific responses to immunization, including tissue-specific differences in oxidative stress and antioxidant defenses.

Keywords: yersiniosis, rainbow trout (Oncorhynchus mykiss), antioxidant defenses, immunization, cardiac tissue, total antioxidant capacity.

SCREENING FOR ANTIMICROBIAL ACTIVITIES OF THE ETHANOLIC EXTRACT DERIVED FROM FICUS MUCUSO WELW. EX FICALHO LEAVES (MORACEAE) AGAINST BACTERIAL FISH PATHOGENS

Abstract. Use of natural products has been considered as an alternative to antibiotics in fish health management to control bacterial infections in aquaculture. Many plants were shown to have potential for being effective herbal drugs against the fish and other aquaculture pathogens. Therefore, the aim of this study was to test the efficacy of ethanolic extract prepared from Ficus mucuso Welw. ex Ficalho leaves against fish pathogens, Aeromonas hydrophila, Citrobacter freundii, Pseudomonas fluorescens, Yersinia ruckeri to evaluate the possible use of this plant in preventing infections caused by these bacteria in aquaculture. The antimicrobial susceptibility testing was done on Muller-Hinton agar by disc diffusion method (Kirby-Bauer disk diffusion susceptibility test protocol). Muller-Hinton agar plates were inoculated with 200 and 400 μL of standardized inoculum (108 CFU/mL) of bacterium and spread with sterile swabs. Aeromonashydrophila (strain E 2/7/15) isolated locally from gills of rainbow trout (Oncorhynchus mykiss Walbaum) and Pseudomonas fluorescens (strain E 1/7/15) isolated locally from internal organs of rainbow trout with clinical features of furunculosis, Citrobacter freundii isolated locally from gills of eel (Anguilla anguillaL.) with clinical features of disease, as well as Yersinia ruckeri collected from clinically healthy fish andfish with clinical symptoms of yersiniosis were used as test organisms. In our study, ethanolic extracts obtained from F. mucuso proved effective against bacteria tested, with 10-15 mm zones of inhibition being observed. F. mucuso demonstrated the highest antibacterial activity against Citrobacter freundii and Pseudomonas fluorescens. Among various bacteria tested, the highest susceptibility for 400 μL of standardized inoculum (108 CFU/mL) of Citrobacter freundii and Pseudomonas fluorescens was noted. Thus, the preliminary screening as say indicated that F. mucuso leaves extract possess a great potential for the therapy of bacterial infections and may be used as a natural antiseptic and antimicrobial agent in aquaculture. Further investigation needs to be focused on isolation and identification of bioactive compoundsfrom F. mucuso, which would be a platform for further pharmacological studies, in vivo tests and practic alapplications in fish health management.

Keywords: Aeromonas hydrophila, Citrobacter freundii, Pseudomonas fluorescens, Yersinia ruckeri, antimicrobial activity, disc diffusion technique, ethanolic extract.

ANTIOXIDANT ACTIVITY OF SOME MOLLUSCS SPECIES FERMENTOLISATES

Abstract. Content of soluble protein, free amino acids and peptides of bivalves fermentolysates was determined. The influence of introducing whey methods on fermentolysates composition was determined. The content of redox-amino acids of mollusks fermentolysates is calculated.
